Mechanism map

Start with the coenzyme. Not the bottle.
Analogy: NAD+ is like a rechargeable coin in a machine shop. Some coins move the assembly line. Some coins get spent on repair crews. When the till runs low, both jobs slow down.
Why people talk about “boosting NAD+”: If pools fall with age or metabolic stress, researchers ask whether raising them helps. A 2013 Cell paper linked falling NAD+ to a “pseudohypoxic” communication state between the nucleus and mitochondria in aging mice. That is biology framing. It is not a shopping list.
Precursor lane — classic mouse files:
- A 2011 Cell Metabolism paper reported NMN helped diet- and age-related metabolic dysfunction in mice and restored NAD+ levels in that design.
- A 2012 Cell Metabolism paper showed NR boosted oxidative metabolism and protected against high-fat-diet obesity in mice.
- A 2016 Cell Metabolism paper reported long-term NMN eased age-linked decline in mice.
- A 2016 Nature Communications paper showed NR is orally available in mice and humans and raises NAD+ metabolome measures.
Reviews in 2015, 2018, and 2021 place NAD+ at the crossroads of energy metabolism, DNA repair, chromatin, senescence, and immune function. They also note age-linked declines across tissues and models.
Human precursor lane — emerging, not a crown:
- 2018: chronic NR was well tolerated and raised NAD+ in healthy middle-aged and older adults.
- 2019: NR changed the aged human skeletal muscle NAD+ metabolome in that study design.
- 2020: oral NMN clinical parameters were studied in healthy men.
- 2021: a randomized Science trial reported NMN increased muscle insulin sensitivity in postmenopausal women with prediabetes.
- 2023: a review mapped what dietary NAD+-boosting compounds do in humans — and what is still open.
Research-vial lane versus precursor lane: A freeze-dried NAD+ research SKU is the coenzyme itself in a bottle. Oral NMN and NR papers ask whether precursors raise NAD+ after digestion and tissue uptake. Related talks. Not interchangeable receipts.
